Agents take queries seriously. And, yes, they really do read them. A query letter is a single-page cover letter meant to elicit an invitation from an agent or publisher to send a sample of your work or even the whole manuscript.
A book proposal--a 20-40 page document that pitches the book's content and marketability, and is supplemented by sample chapers--often comes after the query letter.
